Will the victim know it is a prank right away?

Not likely. Small black marks appearing randomly on the screen look like dead pixels, display glitches, or even actual dirt on the monitor. Most victims will try cleaning their screen or adjusting display settings before they even consider that a program might be causing it. The slow, random appearance makes it very hard to detect.
